Abstract

Abstrak Escherichia coli merupakan bakteri gram negatif aerobik dan anaerobik fakultatif dan merupakan flora normal pada saluran pencernaan yang dapat menjadi patogen pada saat di luar tubuh yang keluar bersamaan dengan tinja. Penyakit yang dapat disebabkan oleh bakteri Escherichia coli adalah diare. Daun sirih hitam (Piper betle var nigra) diketahui mengandung senyawa alkaloid,saponin, flavonoid dan polifenol yang memiliki khasiat sebagai antibakteri. Penelitain ini bertujuan untuk mengetahui aktivitas antibakteri ekstak etanol 96% daun sirih hitam. Metode yang digunakan dalam penelitain ini bersifat eksperimen dengan metode ekstraksi menggunakan pelarut etanol 96%. Konsentasi yang di uji yaitu 20%, 40%, 60%, 80% dan 100% .Bakteri uji yang digunakan dalam penelitian ini yaitu strain murni bakteri Escherichia coli. Uji antibakteri dalam penelitian menggunakan metode difusi cakram.Dengan tujuan untuk mengetahui adanya daya hambat ekstrak etanol 96% daun sirih hitam (Piper betle var nigra) terhadap pertumbuhan Escherichia coli. Kata kunci : Escherichia coli, Uji daya hambat, Ekstrak etanol 96% daun sirih hitam (Piper betle var nigra) Abstract Escherichia coli is a facultative aerobic and anaerobic gram-negative bacteria and is a normal flora in the digestive tract that can become pathogenic when it is outside the body that is excreted with feces. Diarrhea is a disease that can be caused by Escherichia coli bacteria. Black betel leaf is known to contain alkaloids, saponins, flavonoids and polyphenols which have antibacterial properties.This study aims to determine the antibacterial activity of 96% ethanol extract of black betel leaf.The method used in this research is experimental with the extraction method using 96% ethanol as solvent.the concentrations tested were 20% ,40% ,60% ,80% and 100%. The test bacteria used in this study were pure strains of Escherichia coli bacteria. Antibacterial test in this study used the disc diffusion method. With the aim of knowing the inhibition of 96% ethanol extract of black betel leaf on the growth of Escherichia coli bacteria.
