Abstract

ANALISIS KLT-BIOAUTOGRAFI DAN AKTIVITAS ANTIBAKTERI EKSTRAK ETANOL RIMPANG LENGKUAS MERAH (Alpinia purpurata (Vieill) K.Schum) TERHADAP BAKTERI Streptococcus pyogenes Rizki padillah, Tresna Lestari, Taufik Hidayat Program S1 Farmasi, Fakultas Farmasi, Universitas Bakti Tunas Husada Tasikmalaya email : Rizkipadillah625@gmail.com ABSTRACT Various secondary metabolite compounds are contained in the red galangal rhizome (Alpinia purpurata (Vieill) K.schum), which is often used for antibacterial purposes. Pharyngitis is a common disease in Indonesia, experienced by almost everyone and caused by the Streptococcus pyogenes bacteria. The purpose of this study was to investigate the effect of ethanol extract from red galangal rhizomes on the growth of Streptococcus pyogenes bacteria, using diffusion disc with various concentrations of 80%, 60%, 40%, 20% and 10%. The minimum inhibitory concentration (MIC) was found to be at concentrations of 9%, 8%, 7%, 6 %, 5%, 4%, 3%, 2 % and 1 %. Thin layer chromatography monitoring was conducted on extracts with various spot appearances using toluene, acetone and formic acid (5:4:1) as eluent, along with TLC-bioautography contact method. Monitoring resulted in Rf values of flavonoid at 0.53 cm, steroid at 0.56 cm and terpenoid groups at 0.65 cm. Showed a minimum inhibitory concentration value at a concentration of 2%. Bioautography TLC testing produced clear zones at Rf values of 0.53 cm and 0.65cm indicating that they were suspected to be compounds of flavonoid and terpenoid group. ABSTRAK Berbagai senyawa metabolit sekunder terkandung dalam rimpang lengkuas merah (Alpinia purpurata (Vieill) K.schum) sering digunakan untuk antibakteri. Faringitis merupakan penyakit yang cukup banyak terjadi di Indonesia. Dialami hampir semua orang, diakibatkan oleh bakteri Streptococcus pyogenes. Tujuan penelitian ini untuk melihat bagaimana pengaruh ekstrak etanol rimpang lengkuas merah terhadap pertumbuhan bakteri Streptococcus pyogenes. Menggunakan difusi cakram dari berbagai variasi konsentrasi ekstrak 80%, 60%, 40%, 20% dan 10%. Konsentrasi hambat minimum (KHM) yaitu 9%, 8%, 7%, 6%, 5%, 4%, 3%, 2% dan 1 %. Dilakukan pemantauan kromatografi lapis tipis (KLT) terhadap ekstrak dengan berbagai penampak bercak. Toluen, aseton dan asam format (5:4:1) digunakan sebagai eluen dan KLT-bioautografi metode kontak. Pemantauan KLT dihasilkan nilai Rf untuk flavonoid 0,53 cm, Rf steroid 0,56 cm serta golongan terpenoid 0,65 cm. Nilai konsentrasi hambat minimum dengan konsentrasi 2%. Pengujian KLT-bioautografi didapat zona bening pada nilai Rf 0,53 cm dan 0,65 cm diduga merupakan senyawa golongan flavonoid dan golongan terpenoid.
