Abstract
Abstrak Kurkumin berasal dari tanaman kunyit (Curcuma longa. L.) yang memiliki sifat antioksidan yang kuat termasuk kedalam senyawa polifenol. Kurkumin termasuk kedalam BCS kelas IV karena memiliki nilai kelarutan dan biovabilitas yang rendah, oleh karena itu tujuan penelitian ini adalah untuk mengidentifikasi karakterisasi dan aktivitas antioksidan SNEDDS kurkumin. Metode yang digunakan adalah pembuatan nanoemulsi dengan campuran zat aktif kurkumin dan Cinnamon oil digunakan sebagai fase minyak, sedangkan surfaktan yang digunakan adalah tween 80 dan ko-surfaktan adalah PEG 400.Pengujian yang dilakukan pada nanoemulsi yaitu PSA, zeta potensial, pdi dan FTIR, serta uji in vitro drug release dan pengujian aktivitas antioksidan. Hasil karakterisasi dari nanoemulsi kurkumin memiliki ukuran partikel 203,9 ± 2,66 nm, nilai zeta potensial -40.1 ± 0,03 mV, nilai indeks polidispersitas 0,269 ± 0,26, hasil in vitro drug release pada rentang waktu 8 jam mencapai 99% dan dari hasil pengujian FTIR terdapat pergeseran serapan gugus fungsi O-H (alkohol, fenol), C-H (alkana, alkena), C-O (alkohol, eter, asam karboksilat). Nanoemulsi kurkumin memiliki nilai IC50 3,89 ± 0,011 µg/mL dan nilai IC50 kurkumin murni adalah 3,08 ± 0,013 µg/mL. hal ini menunjukan SNEDDS kurkumin memiliki aktivitas antioksidan kedalam golongan sangat kuat, Hasil yang diperoleh mengidentifikasi bahwa nilai aktivitas antioksidan SNEDDS kurkumin berada di bawah 50 µg/mL, yang menunjukkan kekuatan antioksidan yang tinggi. Kata Kunci : Antioksidan, kurkumin, nanoemulsi, SNEDDS Abstrack Curcumin is a polyphenolic compound derived from the turmeric plant (Curcuma longa. L) and has very strong antioxidant activity. However, curcumin is included in BCS class IV because it has low solubility and biovability values, therefore this study aims to determine the characterization and antioxidant activity of curcumin SNEDDS. The method used was the preparation of nanoemulsion with a mixture of active substance curcumin and cinnamon oil as carrier oil, surfactant tween 80 and co-surfactant PEG 400. The tests carried out on nanoemulsion were PSA, zeta potential, pdi and FTIR, as well as in vitro drug release test and antioxidant activity test. The characterization results of curcumin nanoemulsion had a particle size of 203.9 ± 2.66 nm, zeta potential value of -40.1 ± 0.03 mV, polydispersity index value of 0.269 ± 0.26, in vitro drug release results in the span of 8 hours reached 99% and from the results of FTIR testing there was a shift in the absorption of O-H functional groups (alcohols, phenols), C-H (alkanes, alkenes), C-O (alcohols, ethers, carboxylic acids). Curcumin nanoemulsion has an IC50 value of 3.89 ± 0.011 µg/mL and the IC50 value of pure curcumin is 3.08 ± 0.013 µg/mL. this shows that SNEDDS curcumin has antioxidant activity which is classified as very strong, the results obtained show the same results classified into very strong antioxidants which are <50 µg/mL.
