Abstract

Abstrak Keberadaan besi (Fe) dalam kadar tertentu sangat dibutuhkan oleh tubuh manusia, namun dalam kadar yang berlebihan dapat menimbulkan efek racun. Untuk memastikan kadar besi (Fe) dalam tubuh manusia perlu dilakukan pengujian, namun harus meggunakan metode yang telah tervalidasi. Validasi metode analisis merupakan salah satu cara untuk memastikan bahwa suatu metode memberikan hasil yang valid dan sesuai tujuan. Tujuan dilakukan pengujian ini yaitu untuk mengetahui hasil validasi metode pada serum menggunakan alat Spektrofotometri Uv-Vis. Pengujian ini dilakukan dengan parameter validasi meliputi : Linearitas, Batas Deteksi (LOD), Batas Kuantifikasi (LOQ), Akurasi, dan Presisi. Hasil pengujian didapatkan nilai persamaan regresi linear y = 0,0415x + 0,0212 dengan nilai koefisien determinasi (R2 ) 0,9876, akurasi (%Recovery) = 99,26, presisi (%RSD) = 1,778% hasil tersebut menunjukan nilai yang memenuhi standar validitas dengan nilai Batas deteksi (LOD) = 3,70 ppm, dan Batas kuantifikasi (LOQ) = 12,29 ppm. Kata Kunci : Besi (Fe), Spektrofotometri Uv-Vis, Validasi Metode, Linearitas, Batas Deteksi (LOD), Batas Kuantifikasi (LOQ), Akurasi, dan Presisi. Abstract The presence of iron (Fe) in certain levels is needed by the human body, but in excessive levels it can cause toxic effects. To ensure the level of iron (Fe) in the human body, it is necessary to carry out testing, but must use a verified method. Verification of analytical methods is one way to ensure that a method provides valid results and is fit for purpose. The purpose of this test is to find out the results of method verification on serum using a Uv-Vis Spectrophotometry tool. This test is carried out with verification parameters including: Linearity, Limit of Detection (LOD), Limit of Quantification (LOQ), Accuracy, and Precision. The test results obtained the value of the linear regression equation y = 0.0415x + 0.0212 with a coefficient of determination (R2) of 0.9876, Limit of detection (LOD) = 3.70 ppm, Limit of quantification (LOQ) = 12.29 ppm, accuracy (%Recovery) = 99.26, precision (%RSD) = 1.778%. The test results show valid results because they meet the acceptance requirements.
