Abstract

Identifikasi Bakteri Sumber Air Panas di Kabupaten Garut Renita, Rochmanah Suhartati, M.Si. Khusnul, M.Si Program studi DIII Analis Kesehatan Universitas BTH Tasikmalaya Abstrak Sumber air panas diperkirakan dapat menjadi media pertumbuhan bakteri, di kabupaten Garut terdapat sumber air panas yang belum tereksplorasi. Dilakukan penelitian ini dengan tujuan untuk mengidentifikasi bakteri, karakteristik dan aktifitas enzim bakteri yang ditemukan. Metode pada penelitian ini adalah deskriptif kualitataif meliputi karakteristik makroskopik dan mikroskopik menggunakan pewarnaan Gram dan spora serta uji aktifitas enzim bakteri. Hasil penelitian ditemukan 3 isolat dengan kode A1, A2 dan A3. Karakteristik makroskopis yang mirip dan pada pewarnaan Gram semua hasil Gram positif serta ditemukan adanya spora. Isolat yang tumbuh dari sumber air panas di Kabupaten Garut pertumbuhannya lebih subur pada suhu 37oC dibandingkan suhu 50oC. 3 isolat yang tumbuh merupakan tersangka bakteri Bacillus sp kemudian pada uji aktifitas enzim rata-rata memiliki potensi enzimatik amilolitik. Kata kunci : Bakteri Sumber Air Panas, Enzim Amilolitik Abstract Hot springs are thought to be a medium for bacterial growth, in Garut district there are hot springs that have not been explored. This research was conducted with the aim of identifying the bacteria, the characteristics and activity of the bacterial enzymes found. The method in this study was descriptive qualitative covering macroscopis and microscopis. Characteristics using Gram and spore staining as well as bacterial enzyme activity test. The research result found 3 isolates with codes A1, A2 and A3. The macroscopic characteristics were similar and on Gram staining all result were Gram positive and spores were found. Isolates that grew from hot springs in Garut Regency grew more fertile at a temperature of 37oC compared to 50oC. The 3 isolates that grew were suspected to be Bacillus sp bacteria, then on average the enzyme activity test had amylolytic enzymatic potential. Keywords : Hot Spring Bacteria, Amylolytic Enzym
