Abstract

Fawwaz Haifa Program Studi S-1 Farmasi, Universitas Bakti Tunas Husada Abstrak Indonesia negara tropis yang dilintasi garis khatulistiwa terpapar sinar matahari sepanjang tahun. Paparan sinar UVB intens dapat menyebabkan stres oksidatif dan overproduksi melanin yang dikenal sebagai melanogenesis. Melanogenesis adalah mekanisme perlindungan kulit terhadap radiasi UV, namun peningkatan melanin dapat menyebabkan hiperpigmentasi. Penelitian ini bertujuan untuk mengevaluasi aktivitas antimelanogenesis ekstrak angkak (Monascus purpureus) terhadap histopatologi kulit kelinci yang diinduksi sinar UVB. Angkak produk beras terfermentasi, kaya akan flavonoid yang dikenal sebagai penghambat tirosinase, enzim kunci dalam melanogenesis. Ekstraksi angkak dilakukan dengan metode maserasi menggunakan etanol 70%, menghasilkan rendemen 10,1578%. Pengujian mutu ekstrak menunjukkan susut pengeringan 0,6061%, kadar air 0,4635%, dan kadar abu 0,8430%, semuanya memenuhi standar. Penapisan fitokimia mengidentifikasi adanya kuinon, saponin, dan flavonoid. Sediaan salep ekstrak angkak diformulasikan dalam konsentrasi 4%, 8%, dan 16%, menunjukkan karakteristik hasil evaluasi yang baik. Pengujian histopatologi pada kulit kelinci yang diinduksi UVB menunjukkan bahwa sediaan ekstrak angkak, terutama pada konsentrasi 16%, efektif dalam menghambat pembentukan melanin dan mengurangi penebalan epidermis dibandingkan kontrol negatif. Flavonoid dalam angkak berperan sebagai antioksidan dan inihibitor tirosinase, mencegah kerusakan sel akibat ROS dan menekan melanogenesis. Hasil ini mengindikasikan potensi angkak sebagai agen antimelanogenesis. Kata kunci: Angkak, Antimelanogenesis, Penebalan Epidermis, Melanin. Abstract Indonesia a tropical country traversed by the equator is exposed to sunlight throughout the year. Intense UVB radiation can lead to oxidative stress and melanin overproduction, known as melanogenesis. Melanogenesis is a skin protection mechanism against UV radiation, but increased melanin can cause hyperpigmentation. This study aimed to evaluate the antimelanogenesis activity of red yeast rice (Monascus purpureus) extract on the histopathology of rabbit skin induced by UVB light. Red yeast rice, a fermented rice product, is rich in flavonoids, known as tyrosinase inhibitors, a key enzyme in melanogenesis. The red yeast rice extract was obtained by maceration using 70% ethanol, yielding 10.1578%. Quality control tests of the extract showed a drying shrinkage of 0.6061%, water content of 0.4635%, and ash content of 0.8430%, all meeting the specified standards. Phytochemical screening identified the presence of quinones, saponins, and flavonoids. Ointment preparations of the red yeast rice extract were formulated at concentrations of 4%, 8%, and 16%, demonstrating good evaluation characteristics. Histopathological testing on UVB-induced rabbit skin revealed that the red yeast rice extract preparations, particularly at 16% concentration, were effective in inhibiting melanin formation and reducing epidermal thickening compared to the negative control. Flavonoids in red yeast rice act as antioxidants and tyrosinase inhibitors, preventing cell damage from ROS and suppressing melanogenesis. These results indicate the potential of red yeast rice as an antimelanogenesis agent. Keywords: Red Yeast Rice, Antimelanogenesis, Epidermal Thickening, Melanin.
