Abstract

PROGRAM STUDI DIII ANALIS KESEHATAN FAKULTAS ILMU KESEHATAN UNIVERSITAS BAKTI TUNAS HUSADA TASIKMALAYA Karya Tulis Ilmiah, Juli 2025 Ninda Nuraeni Aktivitas Antidiabetes Infusa Daun Sirih Merah (Piper crocatum Ruiz & Pav.) Terhadap Enzim Alfa-Amilase Secara In Vitro Abstrak Diabetes Melitus (DM) merupakan gangguan metabolisme kronis yang ditandai dengan peningkatan kadar glukosa darah. Salah satu pendekatan dalam pengelolaan DM adalah menghambat enzim α-amilase yang berperan dalam pemecahan karbohidrat. Daun Sirih Merah (Piper crocatum Ruiz & Pav.) mengandung senyawa aktif seperti flavonoid, alkaloid, tanin, dan saponin yang berpotensi sebagai inhibitor enzim pencerna karbohidrat. Penelitian ini bertujuan untuk mengetahui aktivitas antidiabetes infusa daun sirih merah secara in vitro terhadap enzim α-amilase dan menentukan nilai IC₅₀. Metode yang digunakan meliputi pembuatan infusa, skrining fitokimia, serta uji aktivitas enzim α-amilase dengan metode DNS dan pembacaan spektrofotometer pada panjang gelombang 540 nm. Hasil penelitian menunjukkan bahwa infusa daun sirih merah mampu menghambat aktivitas enzim α-amilase, dengan persentase inhibisi tertinggi sebesar 74,25% pada konsentrasi 2,55%. Nilai IC₅₀ yang diperoleh sebesar 1,55%, menunjukkan bahwa infusa daun sirih merah berpotensi sebagai agen antidiabetik alami. Kata Kunci: daun sirih merah, α-amilase, antidiabetik, infusa, IC₅₀ Abtract Diabetes Melitus (DM) is a chronic metabolic disorder characterized by increased blood glucose levels. One approach in the management of DM is to inhibit the α-amylase enzyme which plays a role in the breakdown of carbohydrates. Red Betel Leaves (Piper crocatum Ruiz & Pav.) contain active compounds such as flavonoids, alkaloids, tannins, and saponins which have the potential as inhibitors of carbohydrate-digesting enzymes. This study aims to determine the antidiabetic activity of red betel leaf infusion in vitro against the α-amylase enzyme and determine the IC₅₀ value. The methods used include making infusions, phytochemical screening, and testing the activity of the α-amylase enzyme using the DNS method and spectrophotometer readings at a wavelength of 540 nm. The results showed that red betel leaf infusion was able to inhibit the activity of the α-amylase enzyme, with the highest inhibition percentage of 74.25% at a concentration of 2.55%. The IC₅₀ value obtained was 1.55%, indicating that red betel leaf infusion has the potential as a natural antidiabetic agent. Key word: red betel leaf, α-amylase, antidiabetic, infusion, IC₅₀
