Abstract

Rahma Nurazizah DIII Analis Kesehatan/TLM, Universitas Bakti Tunas Husada Tasikmalaya Abstrak Diabetes melitus merupakan penyakit metabolik kronis yang ditandai dengan peningkatan kadar glukosa darah. Salah satu strategi pengelolaan diabetes adalah dengan menghambat enzim α-amilase, yang berperan dalam pemecahan karbohidrat menjadi glukosa. Penelitian ini bertujuan untuk mengetahui aktivitas antidiabetes oleh ekstrak daun sirih merah (Piper crocatum Ruiz & Pav.) asal Manonjaya melalui penghambatan aktivitas enzim α-amilase secara in vitro, serta menentukan konsentrasi yang mampu menghambat 50% aktivitas enzim (IC50). Ekstrak daun sirih merah diproses dengan metode maserasi. Uji penghambatan enzim dilakukan dengan metode DNS, dan pembacaan absorbansi menggunakan spektrofotometri pada panjang gelombang 540 nm. Nilai IC50 yang diperoleh sebesar 10,24% yang termasuk dalam kategori aktivitas penghambatan sangat lemah. Dengan demikian, ekstrak daun sirih merah asal Manonjaya menunjukkan potensi sebagai agen antidiabetes melalui penghambatan aktivitas enzim α-amilase secara in vitro. Kata Kunci: Daun sirih merah, α-amilase, antidiabetes, IC50, penghambatan enzim. Abstract Diabetes mellitus is a chronic metabolic disease characterized by increased blood glucose levels. One strategy for diabetes management is to inhibit the α-amylase enzyme, which plays a role in breaking down carbohydrates into glucose. This study aims to determine the antidiabetic activity of red betel leaf (Piper crocatum Ruiz & Pav.) extract from Manonjaya through the inhibition of α-amylase enzyme activity in vitro, as well as to determine the concentration capable of inhibiting 50% of enzyme activity (IC50). The red betel leaf extract was processed using the maceration method. The enzyme inhibition test was conducted using the DNS method, and absorbance was measured using a spectrophotometer at a wavelength of 540 nm. The IC50 value obtained was 10,24%, which is categorized as very weak inhibitory activity. Thus, the red betel leaf extract from Manonjaya shows potential as an antidiabetic agent through the inhibition of α-amylase enzyme activity in vitro.
