Abstract

Abstrak Tanaman hanjuang (Cordyline fruticosa L.) secara empiris digunakan sebagai obat batuk berdahak namun belum ada bukti secara ilmiah terkait aktivitas mukolitik pada daun hanjuang. Penelitian ini bertujuan untuk menguji mutu simplisia, bobot jenis ekstrak serta efektifitas mukolitik ekstrak etanol 70% (EEHM) dan ekstrak terpurifikasi daun hanjuang merah (EPHM) serta pengaruh proses deffating terhadap aktivitas mukolitik dengan konsentrasi masing-masing (0,5, 1, dan 1,5) %. Hasil pengujian, simplisia daun hanjuang memiliki mutu yang baik. Seluruh konsentrasi EEHM dan EPHM menujukkan aktivitas mukolitik dengan adanya penurunan viskositas mukus usus sapi secara signifikan. Berdasarkan data LSD, EEHM dan EPHM dengan konsentrasi 1 dan 1,5 % memiliki efektivitas yang sebanding dengan asetilsistein 0,1 % sebagai kontrol positif (P>0,05), sedangkan pada konsentrasi 0,5% ekstrak daun hanjuang merah memiliki efektivitas yang lebih baik dibanding kontrol positif (P<0,05) dengan persen penurunan viskositas yang lebih tinggi. Proses purifikasi tidak memberikan pengaruh yang signifikan terhadap aktivitas mukolitik berdasarkan independent sample T-test (P>0,05). Kata kunci: Cordyline fruticosa, Hanjuang Merah, In-vitro, Mukolitik, Mukus usus sapi Abstract Hanjuang (Cordyline fruticosa L.) is empirically used as a cough medicine, but there is no scientific evidence regarding the mucolytic activity of hanjuang leaves. This study aims to evaluate the quality of the crude drug, the specific gravity of the extract, and the mucolytic efficacy of 70% ethanol extract (EEHM) and purified extract of red hanjuang leaves (EPHM), as well as the effect of the defatting process on mucolytic activity at concentrations of 0.5%, 1%, and 1.5%. The results of the testing showed that the hanjuang leaf crude extract has good quality. All concentrations of EEHM and EPHM demonstrated mucolytic activity, with a significant reduction in the viscosity of bovine intestinal mucus. Based on LSD data, EEHM and EPHM at concentrations of 1 and 1.5% had efficacy comparable to acetylcysteine 0.1% as the positive control (P>0.05), while at a concentration of 0.5%, red hanjuang leaf extract had better efficacy than the positive control (P<0.05) with a higher percentage reduction in viscosity.
