Abstract

Abstrak Penyakit Alzheimer merupakan gangguan neurodegeneratif yang berkaitan dengan penurunan fungsi kolinergik karena defisiensi asetilkolin yang disebabkan oleh hiperaktivitas enzim asetilkolinesterase (AChE). Penelitian ini bertujuan untuk mengidentifikasi potensi senyawa bioaktif daun sage (Salvia officinalis L.) sebagai inhibitor AChE melalui pendekatan in silico. Sebanyak 47 senyawa bioaktif diuji terhadap reseptor 4EY7, 6O4W, dan 7E3H menggunakan molecular docking, analisis interaksi, drug scan, ADMET, serta molecular dynamics 100 ns. Hasil menunjukkan bahwa rosmarinic acid merupakan kandidat utama karena memiliki afinitas ikatan yang baik (-127,66 kcal/mol), profil Lipinski dan ADMET relatif mendukung, serta kompleks yang stabil. Beta sitosterol, luteolin-7-O-glucoside, dan apigenin-7-O-glucoside juga berpotensi sebagai kandidat pendukung. Kata Kunci: in silico, asetilkolinesterase, Alzheimer, Salvia officinalis Abstract Alzheimer’s disease is a neurodegenerative disorder associated with cholinergic dysfunction due to acetylcholine deficiency caused by acetylcholinesterase (AChE) hyperactivity. This study identify bioactive compounds sage leaves (Salvia officinalis L.) as potential AChE inhibitors using an in silico approach. A total of 47 bioactive compounds were tested against 4EY7, 6O4W, and 7E3H receptors using molecular docking, interaction analysis, drug scan, ADMET, and 100 ns molecular dynamics. The results showed that rosmarinic acid is the main candidate because it has good binding affinity (-127,66 kcal/mol), favorable Lipinski and ADMET profiles, and stable complex formation. Beta-sitosterol, luteolin-7-O-glucoside, and apigenin-7-O-glucoside also showed potential as supporting candidates. Keywords: in silico, acetylcholinesterase, Alzheimer, Salvia officinalis
