Abstract
Abstrak Hipertensi meningkat secara signifikan dalam skala global, serta dianggap penyebab utama pernyakit kardiovaskular. Berdasarkan penelitian-penelitian sebelumnya, seledri menunjukkan potensi besar sebagai agen antihipertensi. Oleh karena itu, penelitian ini bertujuan untuk mengidentifikasi senyawa aktif dalam daun seledri secara in silico yang dapat berfungsi sebagai penghambat reseptor antihipertensi. Penelitian dilakukan secara in silico dengan molecular docking (PyRx) dan molecular dynamic (Desmond), analisis Lipinski’s Rule Of Five serta pemeriksaan farmakokinetik dan toksisitas (pkcsm). Hasil molecular docking Campesterol mempunyai binding affinity -9,25 kcal/mol dan konstanta inhibisi yang dimilikinya 164,96nM. Visualisasi Campesterol memiliki 12 residu asam amino yang sama dengan ligan alami dan 10 residu yang sama dengan Captopril. Campesterol juga memenuhi parameter uji prediksi farmakokinetik dan toksisitas namun memiliki nilai total clearance terendah dibandingkan dengan senyawa lainnya. .Hasil molecular dynamic senyawa Campesterol menjadi paling unggul dan berpotensi sebagai inhibitor angiotensin convertinf enzme karena mempunyai interaksi yang paling stabil selama proses simulasi. Keywords: Antihipertensi, Angiotensin converting enzyme, celery , Captopril, In-Silico Abstract Hypertension has significantly increased on a global scale and is considered a leading cause of cardiovascular disease. Previous studies have demonstrated that celery possesses strong potential as an antihypertensive agent. This study aims to identify bioactive compounds in celery (Apium graveolens) leaves through an in-silico approach, evaluating their potential as inhibitors of antihypertensive targets. The research employed molecular docking using PyRx and molecular dynamics simulations using Desmond, followed by Lipinski’s Rule of Five assessment and pharmacokinetic and toxicity predictions via pkCSM. Campesterol exhibited a binding affinity of -9.25 kcal/mol and an inhibition constant of 164.96 nM. Visualization revealed that Campesterol shares 12 amino acid residues with the native ligand and 10 residues with the standard drug, Captopril. It also fulfilled pharmacokinetic and toxicity criteria, although it showed the lowest total clearance among the tested compounds. Molecular dynamics simulations confirmed that Campesterol maintained the most stable interactions throughout the 100 ns simulation, supporting its potential as an effective angiotensin-converting enzyme inhibitor.
