Abstract

ABSTRACT Characterized by abnormally high systemic cholesterol, hypercholesterolemia remains a major predisposing factor for cardiovascular conditions. Consequently, botanical therapies are gaining traction as well-tolerated alternative interventions. Leaves of Pandanus amaryllifolius Roxb. are rich in functional phytochemicals—namely flavonoids, polyphenols, alkaloids, and saponins that potentially exert hypocholesterolemic effects. Here, we investigated the lipid-lowering potential of ethanolic P. amaryllifolius leaf extract in hypercholesterolemic Wistar rat models induced by quail egg yolk and propylthiouracil (PTU). The subjects were categorized into normal control, negative control, positive control (simvastatin), and three intervention cohorts receiving extract doses of 10, 20, or 40 mg/kg BW. Serum total cholesterol was measured via the CHOD-PAP method. Compared to the negative control group, the ethanolic extract produced significant dose-dependent cholesterol reductions (p<0.05, One-way ANOVA) of 17.62%, 30.05%, and 41.96% for the 10, 20, and 40 mg/kg BW treatments, respectively. Overall, P. amaryllifolius leaf extract exhibits remarkable antihypercholesterolemic activity, with 40 mg/kg BW established as the most effective dose. Keywords: Pandanus amaryllifolius, hypercholesterolemia,cholesterol, flavonoid, Wistar rats ABSTRAK Sebagai salah satu faktor pemicu gangguan kardiovaskular, hiperkolesterolemia dapat diatasi melalui terapi herbal yang dinilai relatif aman. Daun pandan wangi (Pandanus amaryllifolius Roxb.) memuat metabolit sekunder—termasuk flavonoid, polifenol, alkaloid, dan saponin—yang berpotensi menekan kadar kolesterol. Penelitian ini mengevaluasi daya turun ekstrak etanol daun pandan wangi terhadap kolesterol total pada hewan uji yang diinduksi kombinasi kuning telur puyuh dan PTU. Menggunakan metode CHOD-PAP, evaluasi dilakukan pada kelompok kontrol (normal, negatif, dan positif simvastatin) serta kelompok perlakuan dosis 10, 20, dan 40 mg/kg BB. Hasil analisis One Way ANOVA menunjukkan adanya perbedaan signifikan antar perlakuan (p<0,05), di mana ekstrak mampu mengontrol kadar kolesterol melebihi kelompok kontrol negatif. Efektivitas penurunan kolesterol tertinggi dicapai oleh dosis 40 mg/kg BB (41,96%), disusul dosis 20 mg/kg BB (30,05%), dan dosis 10 mg/kg BB (17,62%).
