Abstract
ABSRTRAK Uji Aktivitas Nefroprotektif Ektrak Etanol Daun Teh-Tehan (Acalypha Siamensis) Terhadap Tikus Putih Jantan Yang Diinduksi Gentamisin Dhea Monika Program Studi S1 Farmasi, Universitas Bakti Tunas Husada Abstrak Kerusakan ginjal akibat penggunaan obat nefrotoksik seperti yang dapat menyebabkan peningkatan stres oksidatif dan kerusakan sel tubulus ginjal. Daun teh-tehan (Acalypha siamensis) diketahui mengandung senyawa antioksidan seperti flavonoid dan fenolik yang berpotensi melindungi ginjal dari kerusakan oksidatif. Penelitian ini bertujuan mengetahui aktivitas nefroprotektif ekstrak etanol daun teh-tehan (Acalypha siamensis) terhadap tikus putih jantan galur Wistar yang diinduksi gentamisin. Penelitian eksperimental ini menggunakan 25 ekor tikus yang dibagi menjadi lima kelompok, yaitu kontrol normal, kontrol negatif, dosis 100 mg/200 gram BB tikus, 200 mg/200 gram BB tikus, dan 400 mg/200 gram BB tikus. Gentamisin diberikan sebanyak 80 mg/kgBB selama 8 hari sebagai penginduksi nefroprotektif. Parameter yang diamati meliputi kadar kreatinin dan BUN. Data dianalisis menggunakan SPSS 32 dengan uji normalitas, uji homogenitas. uji ANOVA dan uji LSD. Hasil penelitian menunjukkan bahwa dosis 200 mg/kgBB memberikan aktivitas nefroprotektif paling optimal dengan persentase aktivitas kreatinin sebesar 77,77% dan ureum sebesar 54,85%, serta kadar kreatinin akhir 0,60 mg/dL dan ureum 38,92 mg/dL yang mendekati kelompok normal (p<0,05). Kata kunci : Acalypha Siamensis, nefroprotektif, kreatinin, BUN. Abstract Kidney damage caused by the use of nephrotoxic drugs may lead to increased oxidative stress and damage to renal tubular cells. Teh-tehan leaves (Acalypha siamensis) are known to contain antioxidant compounds such as flavonoids and phenolics, which have the potential to protect the kidneys from oxidative damage. This study aimed to determine the nephroprotective activity of ethanol extract of teh-tehan leaves (Acalypha siamensis) in male Wistar white rats induced with gentamicin. This experimental study used 25 rats divided into five groups: normal control, negative control, dose of 100 mg/200 g body weight, 200 mg/200 g body weight, and 400 mg/200 g body weight. Gentamicin was administered at a dose of 80 mg/kg body weight for 8 days as a nephrotoxic inducer. The observed parameters included creatinine and blood urea nitrogen (BUN) levels. Data were analyzed using SPSS 32 with normality test, homogeneity test, ANOVA test, and LSD test. The results showed that the dose of 200 mg/ g body weight provided the most optimal nephroprotective activity, with creatinine activity percentage of 77.77% and urea activity percentage of 54.85%, as well as final creatinine levels of 0.60 mg/dL and urea levels of 38.92 mg/dL, which were close to the normal group (p<0.05).
