Abstract
BSTRAK Pemeriksaan Koloni Bakteri Escherichia coli dan coliform Pada Sumber Air di Institusi X dengan Metode Membran Filter Sri Utami Widiawati (Program Studi DIII Analis Kesehatan Universitas Bakti Tunas Husada) Abstrak Air yang digunakan untuk kebutuhan sehari-hari harus memenuhi persyaratan kualitas mikrobiologi agar aman bagi kesehatan. Keberadaan bakteri Escherichia coli dan Coliform dapat menunjukkan adanya pencemaran mikrobiologis pada sumber air. Penelitian ini bertujuan untuk mengetahui jumlah koloni Escherichia coli dan Coliform pada sumber air di Institusi X menggunakan metode membran filter. Penelitian ini merupakan penelitian deskriptif kuantitatif dengan sampel berupa empat sumber air yang berada di lingkungan Institusi X. Pemeriksaan dilakukan menggunakan metode membran filter dan media CompactDry EC, kemudian hasil dinyatakan dalam satuan CFU/100 mL. Hasil penelitian menunjukkan bahwa seluruh sampel positif mengandung Escherichia coli dan Coliform. Jumlah koloni Escherichia coli berkisar antara 3–22 CFU/100 mL, sedangkan jumlah koloni Coliform berkisar antara 13–118 CFU/100 mL. Berdasarkan hasil penelitian, seluruh sumber air yang diperiksa terkontaminasi Escherichia coli dan Coliform sehingga tidak memenuhi persyaratan kualitas mikrobiologi air berdasarkan Peraturan Menteri Kesehatan Republik Indonesia Nomor 2 Tahun 2023. Abstract Water used for daily needs must meet microbiological quality requirements to ensure its safety for human health. The presence of Escherichia coli and Coliform bacteria may indicate microbiological contamination in water sources. This study aimed to determine the number of Escherichia coli and Coliform colonies in water sources at Institution X using the membrane filter method. This research employed a quantitative descriptive design with four water source samples collected from Institution X. The examination was carried out using the membrane filter method and CompactDry EC medium, and the results were expressed in CFU/100 mL. The results showed that all samples were positive for Escherichia coli and Coliform. The number of Escherichia coli colonies ranged from 3–22 CFU/100 mL, while the number of Coliform colonies ranged from 13–118 CFU/100 mL. Based on the results, all examined water sources were contaminated with Escherichia coli and Coliform and therefore did not meet the microbiological water quality requirements according to the Regulation of the Minister of Health of the Republic of Indonesia Number 2 of 2023.
