ABSTRAK UJI DAYA HAMBAT EKSTRAK ETANOL 70% JAMUR LINGZHI (Ganoderma lucidum) TERHADAP PERTUMBUHAN BAKTERI Escherichia coli SECARA IN VITRO Gladys salsabila DIII Analis Kesehatan, Universitas Bakti Tunas Husada Abstrak Infeksi bakteri Escherichia coli masih menjadi salah satu masalah kesehatan yang sering ditemukan, sementara penggunaan antibiotik yang tidak tepat meningkatkan risiko resistensi bakteri. Jamur Lingzhi (Ganoderma lucidum) diketahui mengandung berbagai senyawa bioaktif yang berpotensi sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak etanol 70% jamur Lingzhi terhadap pertumbuhan bakteri Escherichia coli secara in vitro. Penelitian ini dilakukan menggunakan metode difusi cakram (Kirby-Bauer) pada media Mueller Hinton Agar dengan variasi konsentrasi 10, 20, 30, 40, 50, 60, 70, 80, 90, dan 100% dengan empat kali pengulangan, kloramfenikol sebagai kontrol positif dan aquades steril sebagai kontrol negatif. Hasil skrining fitokimia mengindikasikan adanya flavonoid, alkaloid, saponin, tanin, dan triterpenoid dalam ekstrak. Seluruh konsentrasi ekstrak mampu menghambat pertumbuhan Escherichia coli dengan diameter zona hambat berkisar antara 5,075 mm hingga 9,500 mm. Diameter zona hambat tertinggi diperoleh pada konsentrasi 90% sebesar 9,500 mm. Disimpulkan bahwa ekstrak etanol 70% jamur Lingzhi memiliki aktivitas antibakteri terhadap Escherichia coli. Abstract Escherichia coli bacterial infections are still one of the common health problrms, while improper use of antibiotics increases the risk of bacterial resistance. Lingzhi mushrooms (Ganoderma lucidum) are known to contain various bioactive compounds that potentially act as antibacterials. This study aimed to examine the antibacterial activity of 70% ethanol extract of Lingzhi mushroms against the growth of Escherichia coli in vitro. The study was conducted using the disk diffusion method (Kirby-Bauer) on Mueller Hinton Agar media with concentration 10, 20, 30, 40, 50, 60, 70, 80, 90, and 100%, with four repetitions, chloramphenicol as a positive control and sterile distilled water as a negative control. Phytocemical screening results indicated the presence of flavonoids, alkaloids, saponins, and triterpenoids in the extract. All extract concentrations were able to inhibit the growth of Escherichia coli with inhibition zone diameters ranging between 5,075 mm to 9,500 mm. The highest inhibition zone diameters was obtained at 90% concentration, measuring 9,500 mm.
