Abstract
UJI DAYA HAMBAT EKSTRAK DAUN SIRIH MERAH (piper crocatum) TERHADAP PERTUMBUHAN BAKTERI Staphylococcus aureus PADA LUKA DIABETES Salwa Fauziah Taopik, Rianti Nurpalah, Dina Ferdiani D-III Analis Kesehatan, Universitas Bakti Tunas Husada, Tasikmalaya Abstrak Luka diabetes merupakan salah satu komplikasi Diabetes Mellitus yang sering mengalami infeksi bakteri, terutama Staphylococcus aureus. Penggunaan antibiotik yang tidak rasional dapat meningkatkan risiko resistensi bakteri sehingga diperlukan alternatif antibakteri dari bahan alam. Penelitian ini bertujuan untuk mengetahui kemampuan ekstrak daun sirih merah (Piper crocatum) dalam menghambat pertumbuhan Staphylococcus aureus yang diisolasi dari luka diabetes serta menentukan konsentrasi ekstrak yang paling efektif. Penelitian menggunakan metode eksperimental dengan desain uji difusi cakram (Kirby-Bauer). Pengujian dilakukan di Laboratorium Mikrobiologi Universitas Bakti Tunas Husada menggunakan variasi konsentrasi ekstrak 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, dan 100%. Data diperoleh melalui pengukuran diameter zona hambat dan dianalisis secara statistik menggunakan SPSS. Hasil uji fitokimia menunjukkan adanya kandungan alkaloid, flavonoid, tanin, saponin, steroid, quinon, monoterpenoid, dan fenolik pada ekstrak daun sirih merah. Hasil uji menunjukkan Seluruh konsentrasi ekstrak mampu menghambat pertumbuhan Staphylococcus aureus. Diameter zona hambat terbesar diperoleh pada konsentrasi 50% dengan rata-rata 10,225 mm. Analisis statistik menunjukkan adanya perbedaan aktivitas antibakteri yang bermakna antar kelompok perlakuan. Disimpulkan bahwa ekstrak daun sirih merah memiliki aktivitas antibakteri terhadap Staphylococcus aureus dan konsentrasi 50% merupakan konsentrasi yang paling efektif dalam menghambat pertumbuhan bakteri tersebut. Abstract Diabetic wounds are one of the complications of diabetes mellitus that are frequently subject to bacterial infections, particularly by Staphylococcus aureus. Irrational use of antibiotics can increase the risk of bacterial resistance, making it necessary to seek antibacterial alternatives derived from natural sources. This study aimed to determine the ability of red betel leaf extract (Piper crocatum) to inhibit the growth of Staphylococcus aureus isolated from diabetic wounds and to identify the most effective concentration of the extract. The study employed an experimental method using the disk diffusion test (Kirby-Bauer). The testing was conducted at the Microbiology Laboratory of Bakti Tunas Husada University using extract concentrations of 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, and 100%. Data were obtained by measuring the diameter of the inhibition zones and were statistically analyzed using SPSS. The results of the phytochemical analysis indicated the presence of alkaloids, flavonoids, tannins, saponins, steroids, quinones, monoterpenoids, and phenolics in the red betel leaf extract. The test results showed that all extract concentrations were able to inhibit the growth of Staphylococcus aureus. The largest inhibition zone diameter was obtained at a 50% concentration, with an average of 10.225 mm. Statistical analysis revealed significant differences in antibacterial activity among the treatment groups It was concluded that red betel leaf extract exhibits antibacterial activity against Staphylococcus aureus, and a 50% concentration is the most effective at inhibiting the growth of this bacterium.
