Abstract
Abstrak α-Amilase merupakan biomarker yang berperan penting dalam diagnosis gangguan pankreas, namun metode pemeriksaannya umumnya memerlukan reagen khusus dan instrumen yang relatif mahal. Penelitian ini bertujuan menentukan linearitas, batas deteksi (Limit of Detection/LOD), dan batas kuantifikasi (Limit of Quantification/LOQ) α-amilase menggunakan sistem amilum–iodin berbasis spektrofotometri UV-Vis. Pengukuran dilakukan pada panjang gelombang maksimum 620 nm, kemudian dianalisis menggunakan regresi linear serta perhitungan LOD dan LOQ berdasarkan simpangan baku blanko dan kemiringan kurva kalibrasi. Hasil penelitian menunjukkan hubungan linear yang sangat baik dengan persamaan regresi y = 4,7129x + 0,0135 dan R² = 0,9968. Nilai LOD dan LOQ yang diperoleh masing-masing sebesar 0,00070% dan 0,00212%, menunjukkan metode memiliki sensitivitas yang baik dalam mendeteksi α-amilase pada konsentrasi rendah. Sistem amilum–iodin berbasis spektrofotometri UV-Vis berpotensi dikembangkan sebagai dasar biosensor kolorimetri untuk deteksi α-amilase. Abstract Alpha-amylase is an important biomarker for diagnosing pancreatic disorders; however, conventional assays generally require specialized reagents and costly instrumentation. This study aimed to evaluate the linearity, limit of detection (LOD), and limit of quantification (LOQ) of a starch–iodine-based UV–Visible spectrophotometric method for alpha-amylase detection. Measurements were performed at the maximum wavelength of 620 nm, followed by linear regression analysis and LOD/LOQ calculations based on the blank standard deviation and calibration curve slope. The method showed excellent linearity with the regression equation y = 4.7129x + 0.0135 and R² = 0.9968. The LOD and LOQ values were 0.00070% and 0.00212%, respectively, indicating good sensitivity for detecting low concentrations of alpha-amylase. The starch–iodine UV–Visible spectrophotometric method has potential as the basis for future colorimetric biosensor development.
