Abstract

ABSTRAK Potensi Ekstrak Kulit Terong Ungu (Solanum Melongena L.) Sebagai Pewarnaan Aami Pada Sediaan Apus Darah Tepi (SADT) Aura Silvia Maharani, Meti Kusmiati, Annisa Nur Hasanah DIII Analis Kesehatan, Universitas Bakti Tunas Husada, Tasikmalaya Abstrak Pemeriksaan Sediaan Apus Darah Tepi (SADT) merupakan salah satu pemeriksaan hematologi yang digunakan untuk mengamati morfologi eritrosit, leukosit, dan trombosit. Pewarnaan SADT umumnya menggunakan pewarna sintetis seperti Giemsa yang mampu memberikan kontras warna yang baik, namun memiliki kelemahan berupa potensi toksisitas, biaya relatif tinggi, serta menghasilkan limbah yang kurang ramah lingkungan. Oleh karena itu, diperlukan alternatif pewarna alami yang lebih aman dan mudah diperoleh. Kulit terong ungu (Solanum melongena L.) diketahui mengandung pigmen antosianin yang berpotensi digunakan sebagai pewarna alami. Penelitian ini bertujuan untuk mengetahui potensi ekstrak kulit terong ungu sebagai pewarna alami pada SADT, menilai kualitas pewarnaan pada konsentrasi 30%, 40%, dan 50%, menentukan konsentrasi terbaik, serta membandingkan hasilnya dengan pewarna Giemsa. Penelitian ini merupakan penelitian eksperimental laboratorium dengan desain post-test only control design. Ekstrak kulit terong ungu diperoleh melalui metode maserasi menggunakan etanol 96%. Pewarnaan dilakukan pada preparat SADT dengan konsentrasi ekstrak 30%, 40%, dan 50%, serta kontrol positif menggunakan Giemsa. Setiap perlakuan dilakukan enam kali pengulangan. Penilaian dilakukan secara mikroskopis terhadap morfologi eritrosit, leukosit, trombosit, kualitas warna, dan latar belakang preparat menggunakan metode skoring deskriptif. Hasil penelitian menunjukkan bahwa ekstrak kulit terong ungu mengandung senyawa antosianin dan tanin. Seluruh konsentrasi ekstrak mampu mewarnai eritrosit, leukosit, dan trombosit dengan latar belakang preparat yang bersih. Konsentrasi 30% memberikan hasil pewarnaan terbaik dibandingkan konsentrasi 40% dan 50%, dengan skor pengamatan lebih tinggi pada eritrosit dan leukosit. Namun, kualitas pewarnaan yang dihasilkan masih lebih rendah dibandingkan kontrol Giemsa, terutama pada ketajaman warna dan kejelasan morfologi sel. Abstrac Peripheral Blood Smear (PSS) is a hematology test used to observe the morphology of erythrocytes, leukocytes, and platelets. SST staining generally uses synthetic dyes such as Giemsa, which provide good color contrast. However, it has the potential for toxicity, relatively high cost, and produces environmentally unfriendly waste. Therefore, safer and more readily available natural dye alternatives are needed. Purple eggplant (Solanum melongena L.) peel is known to contain anthocyanin pigments, which have the potential to be used as a natural dye. This study aimed to determine the potential of purple eggplant peel extract as a natural dye for PSS, assess the staining quality at concentrations of 30%, 40%, and 50%, determine the best concentration, and compare the results with Giemsa staining.This study was an experimental laboratory study with a post-test only control design. Purple eggplant peel extract was obtained through a maceration method using 96% ethanol. Staining was performed on PSS preparations with extract concentrations of 30%, 40%, and 50%, as well as a positive control using Giemsa. Each treatment was repeated six times. Microscopic assessments were performed on the morphology of erythrocytes, leukocytes, and platelets, as well as color quality, and the background of the slides using a descriptive scoring method.The results showed that purple eggplant peel extract contains anthocyanin and tannin compounds. All extract concentrations were able to stain erythrocytes, leukocytes, and platelets against a clean slide background. The 30% concentration produced the best staining results compared to the 40% and 50% concentrations, with higher observed scores for erythrocytes and leukocytes. However, the resulting staining quality was still lower than the Giemsa control, particularly in color sharpness and clarity of cell morphology.It can be concluded that purple eggplant peel extract has the potential to be used as a natural dye in peripheral blood smear (SADT) because it can stain blood cell components. However, its staining quality cannot match that of Giemsa staining. The 30% concentration was the best concentration in this study.
