Abstract
Biopharmaceutical Classification System (BCS) ibuprofen termasuk kedalam BCS kelas II yaitu memiliki permeabilitas yang tinggi di usus dan memiliki kelarutan yang rendah dalam air. Tujuan dari penelitian ini adalah untuk menciptakan, mengkarakterisasi, meningkatkan sifat mikromeritik, dan meningkatkan laju disolusi ibuprofen dengan penambahan polimer hidrofilik melalui bentuk spherical co-crystal. Metode spherical co-cryztalization anti solvent menggunakan tiga pelarut: etanol (pelarut baik), aqua DM (pelarut buruk), dan diklorometana (cairan pengikat) spherical co-crystal ibuprofen yang diperoleh dikarakterisasi menggunakan instrumen Scanning Electron Microscopy (SEM) dan Hot Stage Microscopy (HSM) menghasilkan fasa kristal baru, transformasi Infra-Red Fourier (FTIR) menghasilkan pergeseran dan tidak ada gugus fungsi baru, Differential Scanning Calorimetry (DSC) menghasilkan penurunan titik leleh, dan Powder X-ray Diffraction (PXRD) menghasilkan puncak baru yang menunjukkan perbedaan difraktogram sinar-X. SEM menghasilkan morfologi yang tidak rata dan bertekstur kasar dan bulat. Berdasarkan distribusi partikel 0,68 mm, ukuran rata-rata partikel diperoleh. Uji kelarutan dan uji disolusi mengalami peningkatan. Kata Kunci: Anti-Solvent, Ibuprofen, Nikotinamid, Spherical co-crystallization Biopharmaceutical Classification System (BCS) ibuprofen belongs to BCS class II which has high permeability in the intestine and has low solubility in water. The purpose of this study was to create, characterize, improve the micromeritic properties, and increase the dissolution rate of ibuprofen with the addition of hydrophilic polymers through spherical co-crystal form. The anti-solvent spherical co-cryztalization method uses three solvents: ethanol (good solvent), aqua DM (bad solvent), and dichloromethane (binding liquid).The obtained ibuprofen spherical co-crystal was characterized using Scanning Electron Microscopy (SEM) and Hot Stage Microscopy (HSM) instruments resulting in a new crystal phase, Fourier transform Infra-Red (FTIR) resulted in shifts and no new functional groups, Differential Scanning Calorimetry (DSC) resulted in a decrease in melting point, and Powder X-ray Diffraction (PXRD) resulted in new peaks showing differences in X-ray diffractograms. SEM produced an uneven morphology and a rough and rounded texture. Based on the particle distribution of 0.68 mm, the average particle size was obtained.
