Abstract
ABSTRAK Gambaran Kadar Ureum Terhadap Kondisi Kelebihan Zat Besi Pada Ginjal Tikus Yang Diinduksi Hemadex Description of Urea Levels in Iron Excess Conditions in the Kidneys of Rats Induced by Hemadex Nasywa Fathia Alkansa, Tanendri Arrizqiyani DIII Analis Kesehatan Universitas Bakti Tunas Husada Abstrak Zat besi merupakan unsur esensial yang berperan dalam metabolisme tubuh, namun dalam jumlah berlebih dapat menimbulkan efek toksik, termasuk kerusakan ginjal melalui mekanisme stres oksidatif. Penelitian ini bertujuan untuk mengetahui gambaran kadar ureum sebagai indikator fungsi ginjal pada tikus putih (Rattus norvegicus) yang diinduksi zat besi dalam bentuk Hemadex. Desain penelitian menggunakan post-test only control group design dengan sampel 12 ekor tikus jantan yang dibagi menjadi kelompok kontrol (tanpa perlakuan) dan kelompok perlakuan dengan pemberian Hemadex dosis tinggi selama 7 hari. Kadar ureum darah diukur menggunakan metode spektrofotometri. Hasil penelitian menunjukkan bahwa kelompok kontrol memiliki kadar ureum rata-rata 39,67 mg/dL, kelompok perlakuan 1 sebesar 34,9 mg/dL, dan kelompok perlakuan 2 sebesar 44,26 mg/dL, dengan satu individu mengalami lonjakan hingga 87,3 mg/dL. Temuan ini menunjukkan bahwa pemberian Hemadex dapat meningkatkan kadar ureum pada dosis tinggi, yang mengindikasikan adanya gangguan fungsi ginjal akibat akumulasi zat besi. Kesimpulannya, kelebihan zat besi berpotensi menimbulkan nefrotoksisitas yang tercermin melalui peningkatan kadar ureum, sehingga pemantauan fungsi ginjal perlu dilakukan pada penggunaan terapi zat besi parenteral. Kata kunci: Ureum, Zat Besi, Hemadex, Ginjal, Rattus norvegicus Abstract Iron is an essential element involved in various metabolic processes; however, excessive amounts may exert toxic effects, including kidney damage through oxidative stress mechanisms. This study aimed to describe the blood urea levels as an indicator of kidney function in white rats (Rattus norvegicus) induced with iron in the form of Hemadex. The research employed a post-test only control group design with 12 male rats, divided into a control group (without treatment) and treatment groups receiving high doses of Hemadex for seven consecutive days. Blood urea levels were measured using spectrophotometry. The results showed that the average urea levels were 39.67 mg/dL in the control group, 34.9 mg/dL in treatment group 1, and 44.26 mg/dL in treatment group 2, with one subject experiencing a significant increase up to 87.3 mg/dL. These findings indicate that Hemadex administration, particularly at high doses, leads to increased urea levels, suggesting impaired kidney function due to iron accumulation. In conclusion, iron overload has the potential to cause nephrotoxicity, as reflected by elevated urea levels, highlighting the importance of monitoring renal function during parenteral iron therapy.
