Abstract

UJI DAYA HAMBAT EKSTRAK ETANOL 70% JAMUR LINGZHI (Ganoderma lucidum) TERHADAP PERTUMBUHAN BAKTERI Klebsiella pneumoniae Virna Noeraeni , Khusnul, M.Si , Dr. Rudy Hidana, M.Pd D-III Analis Kesehatan, Universitas Bakti Tunas Husada, Tasikmalaya Abstrak Infeksi bakteri Klebsiella pneumoniae merupakan masalah kesehatan yang terus meningkat, terutama akibat resistensi antibiotik, sehingga diperlukan alternatif antibakteri dari bahan alam seperti jamur lingzhi (Ganoderma lucidum). Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak etanol 70% jamur lingzhi terhadap pertumbuhan Klebsiella pneumoniae. Pengujian dilakukan dengan metode difusi cakram pada media Mueller Hinton Agar menggunakan variasi konsentrasi ekstrak (10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90% dan 100%) serta kontrol positif (kloramfenikol) dan kontrol negatif (Aquades steril). Hasil penelitian menunjukkan bahwa seluruh konsentrasi ekstrak mampu menghambat pertumbuhan Klebsiella pneumoniae, dengan diameter zona hambat rata-rata masing-masing sebesar 7,15 mm (10%), 6,80 mm (20%), 6,48 mm (30%), 7,08 mm (40%), 7,15 mm (50%), 8,08 mm (60%), 8,30 mm (70%), 7,80 mm (80%), 7,63 mm (90%), dan 8,60 mm (100%). Berdasarkan kriteria efektivitas hambatan, konsentrasi 10-100% menunjukkan aktivitas sedang, kosentrasi 100% memiliki rata-rata paling tinggi. Kontrol positif menghasilkan zona hambat sebesar 19,08 mm, sedangkan kontrol negatif tidak menunjukkan aktivitas antibakteri. Temuan ini mengindikasikan bahwa ekstrak jamur lingzhi mengandung senyawa yang berpotensi sebagai antibakteri terhadap Klebsiella pneumoniae. Studi lanjutan diperlukan untuk mengidentifikasi senyawa aktif secara spesifik dan menguji efektivitasnya secara in vivo. Abstract Klebsiella pneumoniae bacterial infection is a growing health problem, primarily due to antibiotic resistance. Therefore, alternative antibacterial agents from natural sources, such as lingzhi mushrooms (Ganoderma lucidum), are needed. This study aimed to determine the inhibitory effect of a 70% ethanol extract of lingzhi mushrooms on the growth of Klebsiella pneumoniae. The test was conducted using the disc diffusion method on Mueller Hinton Agar using various extract concentrations (10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, and 100%), as well as positive controls (chloramphenicol) and negative controls (sterile distilled water). The results showed that all extract concentrations were able to inhibit the growth of Klebsiella pneumoniae, with average inhibition zone diameters of 7.15 mm (10%), 6.80 mm (20%), 6.48 mm (30%), 7.08 mm (40%), 7.15 mm (50%), 8.08 mm (60%), 8.30 mm (70%), 7.80 mm (80%), 7.63 mm (90%), and 8.60 mm (100%), respectively. Based on the inhibitory effectiveness criteria, concentrations of 10-100% showed moderate activity, with 100% having the highest average. The positive control produced an inhibition zone of 19.08 mm, while the negative control showed no antibacterial activity. These findings indicate that lingzhi mushroom extract contains compounds with antibacterial potential against Klebsiella pneumoniae. Further studies are needed to identify the specific active compounds and test their effectiveness in vivo.
