Abstract
ABSTRACT The food and cosmetic sectors are progressively incorporating natural bioactive compounds, including tea leaves (Camellia sinensis L.) that contain abundant phytochemical constituents. Nevertheless, the dark brown hue of tea leaf ethanol extract restricts its use in aesthetic-oriented applications. This research aimed to assess the acute toxicity profile of decolorized tea leaf ethanol extract powder for potential utilization as a natural active ingredient in food and cosmetic products. The extraction process involved maceration using 96% ethanol, followed by decolorization through n-hexane partitioning, and subsequent drying via spray-drying technique employing maltodextrin and gum arabic as wall materials. Toxicity evaluation was performed using acute oral toxicity testing in mice (Mus musculus) following OECD 423 protocol at doses reaching 5000 mg/kg body weight, along with organ weight analysis and phytochemical characterization. Findings demonstrated that the decolorization procedure successfully eliminated dark pigments while preserving the primary qualitative phytochemical composition. Throughout the 14-day observation period, no mortality or toxicological symptoms were detected, with all test animals showing normal body weight progression and no substantial alterations in vital organ weights. The extract powder exhibited an oral LD₅₀ exceeding 5000 mg/kg body weight, indicating practical non-toxicity and promising potential as a natural active ingredient for food and cosmetic formulation development. ABSTRAK Industri kosmetik dan pangan semakin mengembangkan bahan aktif alami, salah satunya daun teh (Camellia sinensis L.) yang kaya senyawa bioaktif. Namun, ekstrak etanol daun teh berwarna cokelat gelap sehingga membatasi aplikasinya secara estetika. Penelitian ini bertujuan mengevaluasi keamanan akut serbuk ekstrak etanol daun teh terdekolorisasi sebagai bahan aktif alami untuk aplikasi pangan dan kosmetik. Ekstrak diperoleh melalui maserasi menggunakan etanol 96%, kemudian didekolorisasi dengan partisi n-heksana dan dikeringkan menggunakan metode spray drying dengan maltodekstrin dan gom arab. Keamanan dievaluasi melalui uji toksisitas akut oral pada mencit (Mus musculus) sesuai pedoman OECD 423 hingga dosis 5000 mg/kg BB dan penimbangan bobot organ serta skrining fitokimia. Hasil menunjukkan dekolorisasi mampu menghilangkan pigmen gelap tanpa mengubah profil fitokimia utama. Tidak ditemukan mortalitas maupun gejala toksisitas selama 14 hari pengamatan, dan seluruh hewan mengalami peningkatan berat badan serta tidak terjadi nya perubahan berat organ vital hewan uji. Serbuk ekstrak memiliki nilai LD₅₀ oral >5000 mg/kg BB sehingga tergolong praktis tidak toksik dan berpotensi sebagai bahan aktif alami.
