Abstract

ABSTRAK AKTIVITAS ANTIDIABETES FRAKSI METANOL DAUN SIRIH MERAH (piper crocatum Ruiz & Pav.) BERDASARKAN PENGHAMBATAN ENZIM α-GLUKOSIDASE Jenisa Nadia Fadhilah1, Rianti Nurpalah1, Dina Ferdiani1 DIII Analis Kesehatan/TLM, Universitas Bakti Tunas Husada Abstrak Diabtes melitus merupakan penyakit metabolik yang ditandai oleh peningkatan kadar glukosa darah dan memerlukan pengelolaan jangka panjang. Pemanfaatan bahan alam sebagai sumber senyawa antidiabetes terus dikembangkan, salah satunya melalui penghambatan enzim α-glukosidase. Penelitian ini bertujuan untuk mengetahui aktivitas penghambatan enzim α-glukosidase oleh fraksi metanol daun sirih merah (Piper crocatum Ruiz & Pav.). Metode penelitian meliputi ekstraksi, fraksinasi cair-cair, skrining fitokimia, dan uji aktivitas inhibitor α-glukosidase dengan akarbosa sebagai kontrol positif. Pengujian aktivitas inhibitor α-glukosidase menghasilkan nilai persen inhibisi tertinggi sebesar 89,55% dan nilai IC50 sebesar 1,24%. Hasil tersebut menunjukkan bahwa fraksi metanol daun sirih merah memiliki aktivitas penghambatan terhadap enzim α-glukosidase dan berpotensi sebagai sumber bahan alam antidiabetes. Kata Kunci: Daun sirih merah, α-glukosidase, fraksi metanol, inhibisi Abstract Diabetes mellitus is a metabolic disease characterized by elevated blood glucose levels and requires long-term management. The use of natural ingredients as a source of antidiabetic compounds continues to be developed, one of which is through the inhibition of the α-glucosidase enzyme. This study aimed to determine the α-glucosidase inhibitory activity of the methanol fraction of red betel leaves (Piper crocatum Ruiz & Pav.). The research methods included extraction, liquid-liquid fractionation, phytochemical screening and α-glucosidase inhibitor activity test with acarbose as a positive control. The α-glucosidase inhibitor activity test yielded the highest percentage inhibition value of 89,55% and IC50 value of 1,24%. The result indicate that the methanol fraction of red betel leaves has inhibitory activity afaints the α-glucosidase enzyme and has potential as a source of natural antidiabetic ingredients.
