Abstract
Penyakit diabetes melitus dapat menyebabkan komplikasi kronis berupa luka diabetes yang rentan terinfeksi oleh bakteri, salah satunya Staphylococcus aureus. Peningkatan kasus resistensi antibiotik memerlukan adanya eksplorasi bahan alam sebagai alternatif pengobatan. Jamur lingzhi mengandung berbagai senyawa metabolit sekunder yang berpotensi sebagai antibakteri. Tujuan penelitian ini untuk mengetahui efektivitas ekstrak etanol 70% dalam menghambat pertumbuhan bakteri Staphylococcus aureus dari luka diabetes. Jamur lingzhi diekstraksi menggunakan metode maserari dengan pelarut etanol 70%. Aktivitas antibakteri terhadap Staphylococcus aureus diuji menggunakan metode difusi cakram (Kirby-Bauer) dengan empat kali pengulangan pada sepuluh variasi konsentrasi ekstrak, yaitu 10–100%. Hasil uji fitokimia menunjukkan bahwa ekstrak mengandung flavonoid, tanin, saponin, alkaloid, dan triterpenoid yang di ketahui sebagai senyawa antibakteri. Hasil pengujian menunjukkan seluruh konsentrasi ekstrak menghasilkan aktivitas antibakteri terhadap Staphylococcus aureus. Rata- rata diameter zona hambat yang terbentuk berada pada 5,88-9,98 mm dan dikategorikan sedang. Aktivitas penghambatan paling tinggi pada konsentrasi 60% dengan rata-rata diameter sebesar 9,98 mm. hasil uji statistik menggunakan uji Kruskal-Wallis menunjukkan adanya perbedaan antar variasi konsentrasi. Kesimpulannya ekstrak etanol 70% jamur lingzhi (Ganoderma lucidum) memiliki aktivitas antibakteri terhadap Staphylococcus aureus dengan aktvitas optimum pada konsentrasi 60% dan berpotensi dikembangkan sebagai alternatif terapi alami pada infeksi luka diabetes. Abstract Diabetes can cause chronic issues like diabetic wounds, which are easily infected by bacteria such as Staphylococcus aureus. With antibiotic resistance on the rise, natural remedies are being looked at as alternative treatments. Lingzhi mushrooms have a variety of secondary metabolites that might work as antibacterials. This study looks at how well a 70% ethanol extract can stop the growth of Staphylococcus aureus in diabetic wounds. The Lingzhi mushrooms were extracted using the maceration method with 70% ethanol. Their antibacterial activity against Staphylococcus aureus was tested using the disk diffusion method (Kirby-Bauer), with four repeats across ten extract concentrations from 10% - 100%. Phytochemical tests showed that the extract contains flavonoids, tannins, saponins, alkaloids, and triterpenoids, which are known as antibacterial compounds. The test results showed that all extract concentrations exhibited antibacterial activity against Staphylococcus aureus. The average diameter of the inhibition zones ranged from 5.88 to 9.98 mm, which is categorized as moderate. The highest inhibitory activity was observed at a 60% concentration, with an average diameter of 9.98 mm. Statistical testing using the Kruskal-Wallis test showed differences between the concentration variations. In conclusion, 70% ethanol extract of lingzhi mushrooms (Ganoderma lucidum) has antibacterial activity against Staphylococcus aureus, with optimum activity at 60% concentration, and has potential to be developed as an alternative natural therapy for diabetic wound infections.
